All about Oocyte/Embryos Vitrification

Vitrification can be defined as the transition of water directly from the liquid phase into an amorphous phase or glass, while avoiding the formation of crystalline ice as a unique method of freezing. 

Embryos and eggs can now be successfully frozen and thawed better than ever. Until recently, embryo and egg freezing (also called cryopreservation) were accomplished with a technology known as “slow freezing.” This has recently been replaced with a more sophisticated technology called vitrification, which is generally felt to be a superior process for cryopreservation.

For a long time, the method of slow freezing was the main method of cryopreservation of embryos, as the vitrification, the rapid cooling method, was considered as toxic due to high concentration.

Cryoprotectants in the Cryo solution.

Long-term embryo storage is in liquid nitrogen, at a temperature of –320ºF (-196ºC).
Cryopreserved embryos are replaced during either a natural menstrual cycle or a hormonally controlled cycle. Considerable care is taken to minimize the possibility of damage caused by cryopreservation. Depending on the embryo stage at the time of freezing, between 60 and 90 % survive the freeze/thaw process. The pregnancy rate after transfer of these embryos is similar to that of fresh embryos.

Extended periods of storage in liquid nitrogen have no apparent affect on their viability. Embryos thawed after several years of cryostorage fare as well as those frozen for only one or two months. Since 1983, the cryopreservation procedure has resulted in the birth of thousands of babies worldwide. With this extensive experience, there have been no reports of any increase in birth defects as a result of cryopreservation.
However, the number of researches that were conducted using advanced cryoprotocols showed that the usage of the vitrification method on the different stages of embryo development gives the same or even better results than the method of slow freezing.

In vitrification it is excluded:

  • the danger of physical and chemical damage caused by intercellular and intracellular crystallization;
  • the toxic effect of cryoprotectants, which correlates with the concentration of cryoprotectant, the time and temperature of the embryo exposure in this solution.

  • Fertility preservation in case of  oncological diseases and surgical interventions;
  • Lack of partner at the time of fertilization of oocytes;

How is the vitrification provided?

  • Freezing occurs in special programmable devices that support the cooling rate and the required temperature schedule;
  • Duration - 1,5-2 hours.

Conditions of oocytes storage in Ukraine. 

We are often asked about the expiration date of cryopreserved material, for how many it could stay in the frozen state, and whether this affects the quality of the material. Among embryologists around the world it is commonly accepted that the term does not affect the quality of the cryomaterial, most importantly, the conditions of storage should not be violated. 

At Intersono there is a large and unique cryo repository, where more than 1000 carriers with biomaterials of patients are stored. It is equipped with special sensors,  that react to change of the temperature, if there is a failure, the embryologists come with a beep and there is also a backup power supply. Cryopreservation and safe storage conditions are monitored 24/7. All biomaterials are carefully labeled and contained in special carriers, also called "straws" or "vials" and stored in cryopreservation in special containers of "duars" in which there is liquid nitrogen.

We got surplus embryos  for the most part in the IVF cycles , of which only two or three of them are used for embryo transfer. Therefore, at the request of a couple, we can preserve embryos of good quality in the cryogenic solution so that the woman could use them later (if the pregnancy did not occur at the first embryo transfer or if the woman is getting pregnant, but she wants to become a mother in the future). It happens that our patients already have babies, then they can donate their embryos to other patients. 

For each kind of material (sperm, eggs, embryos) there is a separate freezing technique. We have created an electronic database, which contains all the information of the freezing of the material for each patient. Also, with each patient, we personally will be signed a statement (consent, developed by the Ministry of Health of Ukraine) about the cryopreservation of biomaterial, which specifies the term of storage.

What is the fee for cryopreservation and oocyte storage?

What if  I want to discontinue oocytes/embryos storage? 

Should you ever decide to discontinue storage, we will provide you with a consent form that, when signed personally, will give us permission to stop your storage. We are unable to discontinue your storage without a current completed consent form signed by both partners. Please contact our team to discuss your options and to obtain the proper consent form.

Your IVF coordinator will kindly provide you with full and detailed information in terms of number and quality of frozen eggs/embryos for you. Please advise us for how long would you like to store your biomaterial at our clinic.You will be in constant communication with your personal coordinator to do nor skip a time you should prolonge storage( once per month, per 6 or 12 months).
